Fırat ve Dicle Nehri’nde Yaşayan Barbus grypus (Heckel, 1843) Populasyonlarında Genetik Çeşitliliğin mtDNA COI Gen Dizileri Kullanılarak Belirlenmesi

Abstract

Barbus grypus (Heckel, 1843) Fırat ve Dicle nehir sistemlerinde yaşayan ve
ekonomik önemi olan endemik balık türlerden biridir. Bu çalışmada,
B. grypus
populasyonlarında
mitokondriyal DNA sitokrom c oksidaz altünite I (mtDNA COI) gen bölgesinin dizi
analizlerine dayalı genetik çeşitlilik belirlenmiştir.
İki populasyondan toplam 36 örnek alınıp iki değişken bölge ve üç
haplotip tespit edilmiştir. Ortalama haplotip (h) ve nükleotit çeşitliliği (π)
sırasıyla 0,246 ve 0,00045 olarak hesaplanmıştır.
Nötralite testleri sonucunda tüm değerler
istatistiksel olarak önemsiz bulunmuştur (p>0.10).
Yapılan bu çalışmadaki sonuçlar
B. grypus türü için ilk kez elde
edilmiş verilerdir. mtDNA COI gen bölgesi için belirlenen haplotipler literatür
açısından yeni sonuçlar olup, bu türün genetik çeşitliliği açısından önemli bir
veri seti oluşturmuştur.

Determination of Genetic Diversity in Barbus grypus Heckel, 1843 Populations by mtDNA COI Gene Sequences

Abstract

Barbus grypus (Heckel,
1843) living in the Euphrates and Tigris river system, is one of the endemic
fish species and has economic importance. In this study, genetic diversity of
B. grypus populations was detected based
on mitochondrial DNA cytochrome c oxidase subunit I (mtDNA COI) gene sequences.
Three haplotypes and two polymorphic sites were detected in totally 36
individuals sampled from two populations.
The average haplotype diversity (h) and nucleotide diversity (π)
were calculated
0,246 and 0,00045 respectively. The result of
Neutrality tests as statistically was insignificant (p>0.10). The results
obtained by this study performed firstly for
Barbus grypus species. Determined haplotypes for mtDNA COI gene
region are new results for literature, in terms of genetic diversity of this
species is an important data set.
